作用机制
体外电生理研究和体内神经化学研究表明，伐尼克兰（Varenicline）可与 α4β2 型神经元烟碱型乙酰胆碱受体结合，并刺激受体介导的活性，但其刺激水平显著低于尼古丁。伐尼克兰能够阻断尼古丁对 α4β2 受体的激活作用，从而抑制中枢神经系统中脑边缘多巴胺通路的刺激，该通路被认为是吸烟产生强化效应和奖赏体验的神经机制基础。
伐尼克兰具有高度选择性，其与 α4β2 受体的结合能力显著强于其他常见的烟碱型受体（对 α3β4 受体的选择性高于 500 倍，对 α7 受体高于 3,500 倍，对 α1βγδ 受体高于 20,000 倍），并且对非烟碱型受体和转运体的亲和力也高出 2,000 倍以上。此外，伐尼克兰还以中等亲和力（Ki = 350 nM）与 5-HT3 受体结合。
